[FAQ] TurboMice™ Tetraploid Complementation & Custom Gene-Edited Mouse Models

  1. Technical Principles

Q1: What exactly is tetraploid (4N) complementation? How is it fundamentally different from traditional zygote injection and standard ES cell targeting?
A: Tetraploid complementation is an advanced mouse generation technology. Tetraploid embryos only develop into extraembryonic tissues (placenta, yolk sac), while the entire embryo body is 100% derived from pre-edited, validated embryonic stem cells (ESCs). In contrast, traditional CRISPR zygote injection produces mosaic F0 mice, and standard blastocyst ES injection produces chimeric F0 mice — both require multiple generations of breeding to obtain stable, fully edited strains.

Q2: Why are TurboMice™ F0 mice 100% ESC-derived with zero chimerism, while traditional CRISPR methods produce mosaicism?
A: In conventional CRISPR, editing occurs randomly in individual blastomeres after fertilization, resulting in mixed genotypes within one animal (mosaicism). With our 4N platform, every cell of the resulting mouse originates from a single, fully sequenced and confirmed ESC clone. There is no chimerism and no allele segregation from the F0 generation onward.

Q3: Tetraploid embryos are known to have weak viability. Will this affect the health and fertility of the final mice?
A: No. The tetraploid component contributes only to placental and extraembryonic support tissues; all somatic and germ cells of the born mouse come from normal diploid ESCs. The delivered mice are physiologically normal, fertile, and have a stable genetic background equivalent to conventionally derived strains.

Q4: What model types can 4N technology generate? Are there any limitations?
A: TurboMice™ supports knockout (KO), knock-in (KI), point mutation, Flox, full/partial humanization, multi-gene editing, and large-fragment insertion models. It is especially powerful for complex and large-fragment humanization models that are difficult to achieve via traditional CRISPR. It is compatible with most common inbred and outbred backgrounds.

Q5: Which mouse backgrounds are available? Can we use our own existing ESC lines?
A: We support mainstream backgrounds including C57BL/6N, C57BL/6J, and ICR. We also accept customer-provided qualified ESC lines for 4N complementation, subject to a feasibility review.

  1. Timeline & Delivery

Q1: What is the total project timeline, from ESC engineering to study-ready F0 mice?
A: For a full custom project (gene editing + ESC validation + 4N complementation), we deliver homozygous study-ready F0 mice in 2–4 months. If validated ESCs are already available, mice can be generated in as little as 45 days.

Q2: What genotype do you deliver? Do we receive homozygous mice directly, without further breeding?
A: Yes. We deliver 100% homozygous (e.g., KO/KO, KI/KI, Flox/Flox) F0 mice directly. No F1/F2 intercrossing, no breeding selection, and no waiting for germline transmission — the animals are ready for experiments immediately upon arrival.

Q3: How many F0 mice can be produced in one batch? Can you provide large cohorts of age-matched animals?
A: We can deliver scalable, uniformly age-matched cohorts according to your experimental needs. Since all mice are generated in parallel from the same ESC clone via 4N complementation, we can produce large numbers of genetically identical, same-age mice in a single batch — a major advantage over serial breeding.

  1. Quality & Validation

Q1: What genotyping and quality validation do you provide?
A: We provide PCR genotyping, Sanger sequencing across the targeted locus, and ESC karyotyping reports. All delivered mice come with full genotype confirmation and health documentation.

Q2: Is there an off-target risk? How does it compare to traditional CRISPR zygote injection?
A: Off-target risk is significantly lower. All editing and validation are performed at the ESC level before mouse generation; only confirmed, on-target, correctly edited clones are used for 4N complementation. This eliminates the random off-target and mosaic issues inherent to one-cell embryo injection.

Q3: What pathogen status do the mice have? Can they be directly imported into our SPF facility?
A: All mice are produced and housed under SPF conditions with routine health monitoring. We provide full health reports and can support institutional import requirements, including health certificates and quarantine documentation.

Q4: Is phenotype stable? Is germline transmission guaranteed?
A: Because every mouse is 100% derived from the same validated ESC clone, phenotypes are highly consistent and reproducible across cohorts. Germline transmission is inherent to the technology — germ cells are also fully ESC-derived, so stable transmission is guaranteed.

  1. Breeding & Strain Rescue

Q1: Our existing mouse line has very poor breeding performance or low fertility. Can 4N technology rescue and expand the colony?
A: Yes, this is one of our strongest.
